The tolerance of Persian clover is assessed as the ability to maintain biomass and growth rates under conditions of salinity, waterlogging, or moisture deficiency. It is a key criterion for selecting cultivars adapted to challenging soil and climatic conditions.

Physiological markers are also employed, including analysis of the Na+ and K+ ion ratio in tissues, changes in osmotic potential, and the activity of antioxidant enzymes such as peroxidase and catalase.
In agronomic practice, the species is characterized by high resistance to temporary waterlogging, which allows it to be cultivated on heavy clay soils. Persian clover shows moderate tolerance to salinity, outperforming subterranean clover in this regard, and possesses greater drought resistance compared to berseem clover, although its productivity decreases under moisture deficiency.
