For successful hybridization, maternal A-lines must possess 100% pollen and spikelet sterility, while paternal R-lines must have the ability to restore F1 hybrid fertility to above 90%.
The cultivation technology requires maternal forms to possess specific traits, such as stigma exertion and timely spikelet opening for cross-pollination. Line purity control is carried out using qPCR analysis of mitochondrial markers to exclude B-line impurities, with sterility stability potentially depending on temperature and humidity conditions during the microsporogenesis phase.
