The variety belongs to types with high shoot vigor. The leaves are medium or large, three- or five-lobed, wedge-shaped, with pubescence on the underside. The clusters are medium or large, elongated-conical in shape, medium-dense or loose. The berries are medium, round or ellipsoidal, with thin skin of a golden-yellow color.

Glera is a semi-aromatic variety with high acidity and medium sugar content; the aroma of the wines is dominated by notes of green apple, pear, white flowers, and citrus. The variety has moderate resistance to downy mildew and powdery mildew and is sensitive to drought. Yield is stable, however, it requires crop regulation due to a tendency for ovary and berry shedding.

In literature, the variety is often considered as a group of similar varieties or biotypes, among which the most common are Glera Tondo, Glera Lunga, and Glera Nostrana.
