Citrus species exhibit variable levels of ploidy, with the majority of commercial cultivars being diploid (2n = 2x = 18). The ploidy level significantly influences resistance to abiotic stresses, leaf morphology, and fruit quality characteristics.

For most citrus species, a diploid chromosome set is standard; however, triploid, tetraploid, hexaploid, and aneuploid forms also occur within the Citrus genus. To assess ploidy, cytogenetic analysis of root tips or young shoots is employed, as well as flow cytometry, which allows for the determination of genome size.

The use of polyploid forms, particularly tetraploid rootstocks, helps increase plant adaptability to nutrient deficiency and soil salinity by activating antioxidant systems. Changes in ploidy level affect stomatal apparatus morphology, transpiration rate, and the biochemical composition of essential oils in fruits, making polyploidization an important tool in the breeding of new cultivars.
